They beat Ireland by seven wickets in Dehradun, with almost two days to spare.
Chasing 147 in their fourth innings, Rahmat Shah and Ihsanullah both made half-centuries to guide Afghanistan to their historic maiden win.
Earlier in the match all-rounder Rashid Khan picked up the nation's first five-wicket haul in Tests.
